Overview

TLC-ART 101 is a long-acting, injectable combination antiretroviral therapy (ART) formulated as a drug-combination nanoparticle (DCNP). It co-formulates three small molecule drugs: the protease inhibitors **lopinavir** and **ritonavir**, and the nucleoside reverse transcriptase inhibitor **tenofovir**. Specifically designed for subcutaneous administration, the formulation utilizes a lymphatic-targeting platform to sequester drugs in lymph node depots. This mechanism provides sustained drug release into both the plasma and HIV-target cells, such as peripheral blood mononuclear cells (PBMCs) and lymph node mononuclear cells, for several weeks following a single injection. The platform aims to reduce pill burden and overcome the pharmacokinetic limitations of oral ART, such as the heavy reliance on ritonavir for metabolic boosting.
