Drug intelligence / Profile preview
TLC19 (also known as ISPM19) is an investigational liposomal formulation of hydroxychloroquine (HCQ) developed by Taiwan Liposome Company (TLC) and its subsidiary InspirMed for the treatment and prophylaxis of COVID-19. Utilizing TLC's proprietary LipAD™ lipid-assembled drug delivery platform, the therapy is administered via a portable vibrating mesh nebulizer to deliver the drug directly to the respiratory tract and lungs. This targeted approach aims to achieve high local concentrations—demonstrating 30-fold increased lung exposure in preclinical studies compared to oral HCQ—while using only about 1% of the standard oral dose, thereby potentially avoiding systemic toxicities such as cardiotoxicity. The mechanism of action involves interfering with viral entry and replication by targeting the angiotensin-converting enzyme 2 (ACE2) receptor and modulating endosomal pH.
