Overview

TLD-1433 is a ruthenium-based coordination complex and photosensitizer developed for use in photodynamic therapy (PDT), primarily targeting non-muscle invasive bladder cancer (NMIBC). Upon light activation—optimally in the green spectrum around 520 nm or with red/NIR light—TLD-1433 generates cytotoxic singlet oxygen and reactive oxygen species, leading to efficient cancer cell death. The drug is preferentially absorbed by malignant cells due to their higher expression of transferrin receptor, minimizing toxicity to healthy tissue. In the absence of light ("dark" conditions), TLD-1433 exhibits very low cytotoxicity. The compound has completed Phase 1b clinical trials and is currently in Phase 2 development for NMIBC. It was developed by Theralase Technologies in collaboration with University Health Network[1][2][5][6][7][8].
