The TLPLDC vaccine (tumor lysate, particle-loaded, dendritic cell vaccine) is a personalized therapeutic cancer vaccine designed to prevent recurrence in patients with solid tumors, particularly high-risk melanoma. The process involves isolating autologous dendritic cells from a patient's blood and loading them ex vivo with yeast cell wall particles containing autologous tumor lysate derived from the patient’s own tumor. This approach leverages the immunogenic properties of yeast particles to enhance antigen presentation by dendritic cells. Once administered intradermally, the TLPLDC vaccine delivers a broad repertoire of patient-specific tumor antigens to the immune system, stimulating both innate and adaptive immune responses—most notably activating T cells to recognize and destroy residual or recurring cancer cells. Clinical trials have demonstrated safety and potential efficacy in various solid tumors; most advanced development has been in stage III/IV melanoma[1][2][4][5][8].
