TLPO vaccine is a personalized, acellular cancer vaccine made by loading yeast cell wall particles (YCWP) with autologous tumor lysate (TL) from a patient’s own tumor, then capping the particles with silicate. Unlike the related TLPLDC vaccine, which requires ex vivo loading of dendritic cells, the TLPO vaccine is injected directly and relies on in vivo uptake by dendritic cells, leading to T-cell–mediated anti-tumor immune responses. This method seeks to improve ease of manufacture, reduce cost, and enable direct logistics for personalized immunotherapy. Its primary indication has been in the prevention of recurrence in high-risk, resected stage III and IV melanoma, with ongoing basket trials in other solid tumors. Phase IIb trials demonstrated safety and equivalence in outcomes to TLPLDC, with further Phase III studies planned[1][3][7].
