TLR4-IN-C34 is a small molecule drug that functions as a potent and selective antagonist of Toll-like receptor 4 (TLR4). It is a 2-acetamidopyranoside that inhibits TLR4 signaling in enterocytes and macrophages. By blocking TLR4, it attenuates TLR4-mediated cytokine release and exhibits anti-inflammatory effects. It has been studied in animal models for conditions like endotoxemia and necrotizing enterocolitis, where it reduces systemic inflammation and decreases the expression of inflammatory markers like TNFα and iNOS.
