Overview

The tlr4 polyclonal antibody is a rabbit-derived neutralizing antibody targeting Toll-like receptor 4 (TLR4). It was developed to address the aberrant overactivation of TLR pathways observed in hematopoietic stem cells of patients with Myelodysplastic Syndrome (MDS) and Acute Myeloid Leukemia (AML). By neutralizing TLR4 function, the antibody inhibits downstream signaling pathways, including lipopolysaccharide (LPS)-induced NF-κB activation and the phosphorylation of extracellular signal-regulated kinase (ERK) mitogen-activated protein (MAP) kinase. Preclinical studies using MDS and AML patient samples have demonstrated that treatment with this antibody promotes erythroid and myeloid differentiation, potentially relieving the differentiation block characteristic of these diseases.
