TLX300-CDx is a radiolabeled monoclonal antibody diagnostic agent developed for use in positron emission tomography (PET) imaging. It consists of olaratumab—a human monoclonal antibody that targets platelet-derived growth factor receptor alpha (PDGFRα)—conjugated to the radioisotope zirconium-89 (^89Zr). PDGFRα is frequently overexpressed in soft tissue sarcoma (STS), making it a rational target for both diagnostic and potential therapeutic applications. The primary mechanism involves binding to PDGFRα-expressing tumor cells and enabling PET-based visualization of tumors for patient selection and dosimetry prior to targeted radionuclide therapy. The agent is being evaluated in first-in-human Phase 1 trials as part of a theranostic approach to optimize subsequent therapeutic strategies for advanced or metastatic STS[1][5][8].
