Overview

TLY012 is a PEGylated recombinant human Tumor Necrosis Factor-Related Apoptosis-Inducing Ligand (TRAIL) protein engineered for enhanced potency, stability, and extended half-life. It acts as an agonist of the TRAIL receptor 2 (Death Receptor 5/DR5), selectively targeting and eradicating myofibroblasts in fibrotic tissues. By binding to DR5 on activated fibroblasts (myofibroblasts), it shuts down fibrotic pathways and reverses established fibrosis in preclinical models of liver, skin, and pancreatic fibrosis. The drug is being developed primarily for systemic sclerosis (scleroderma), chronic pancreatitis, liver fibrosis/cirrhosis, and other fibrotic diseases. It has also shown potential anti-tumor effects by promoting apoptosis in cancer cells with upregulated DR5 expression[1][3][4][6][7].
