TM123 is a recombinant antibody-based targeting module (TM) designed for use with the UniCAR (Universal Chimeric Antigen Receptor) T-cell platform. It consists of a single-chain variable fragment (scFv) specific for CD123 (interleukin-3 receptor alpha chain) fused to a peptide epitope recognized by UniCAR-T cells. Unlike conventional CAR-T cells that directly bind to tumor antigens, UniCAR-T cells are inert until they are cross-linked to target cells by a soluble targeting module like TM123. This switchable system allows for precise control over T-cell activity and duration, potentially reducing toxicities such as cytokine release syndrome and on-target/off-tumor effects. TM123 is primarily being developed for the treatment of CD123-positive hematologic malignancies, including acute myeloid leukemia (AML).
