Overview

TM4SF1-directed chimeric antigen receptor T cells (CART-TM4SF1) are an investigational cell therapy designed to target Transmembrane 4 L Six Family Member 1 (TM4SF1), a protein highly expressed in various solid tumors, particularly those of the digestive system. TM4SF1 plays a role in cell proliferation, migration, and angiogenesis. These CAR-T cells are typically engineered (some versions as 3rd generation) to recognize TM4SF1-expressing tumor cells and induce targeted cell death. To enhance safety, some constructs incorporate an EGFR-based safety switch, allowing for the depletion of the CAR-T cells if severe toxicity occurs. Clinical evaluation is ongoing for indications including pancreatic, colorectal, gastric, liver, ovarian, and biliary tract cancers.
