Overview

TM4SF1-MYC siRNA conjugate is an antibody-RNA conjugate (ARC) being developed by Tavotek Lab for the treatment of various solid tumors. The therapeutic consists of a monoclonal antibody targeting Transmembrane-4-L-six-family member-1 (TM4SF1) conjugated to a small interfering RNA (siRNA) designed to silence the MYC oncogene. TM4SF1 is a tetraspanin protein frequently overexpressed in gastric, lung, pancreatic, and breast cancers, where it facilitates tumor progression, migration, and angiogenesis. By leveraging the high affinity of the anti-TM4SF1 antibody, the conjugate selectively delivers the MYC siRNA into cancer cells. Once internalized, the siRNA mediates the knockdown of MYC protein, a key driver of cell proliferation and survival in many cancers. Preclinical data presented at AACR 2025 demonstrated that the conjugate is stable and effectively inhibits tumor cell growth in MYC-dependent lung, pancreatic, and gastric cancer models. The program is currently undergoing IND-enabling studies.
