TM4SF1-positive chimeric antigen receptor T-cell therapy (also known as CART-TM4SF1) is an investigational autologous CAR-T cell therapy developed by Changhai Hospital of Shanghai in collaboration with Shanghai Ultra-T Immune Therapeutics. The therapy targets Transmembrane 4 L Six Family Member 1 (TM4SF1), a plasma membrane glycoprotein that is highly expressed in various digestive system cancers, including pancreatic, colorectal, gastric, and liver cancers, as well as on the vascular endothelial cells lining tumor blood vessels. By targeting TM4SF1, the engineered T-cells aim to eliminate tumor cells and inhibit tumor angiogenesis. The CAR construct includes an epidermal growth factor receptor (EGFR)-based safety switch to allow for the selective depletion of the CAR-T cells if necessary. It is currently being evaluated in Phase 1 clinical trials for patients with recurrent or refractory advanced TM4SF1-positive digestive system tumors.
