TM4SF1-targeting ADC is an antibody-drug conjugate (ADC) directed against Transmembrane 4 L six family 1 (TM4SF1), a protein belonging to the transmembrane 4 L six family. TM4SF1 is highly expressed in various epithelial-derived malignancies, including lung, pancreatic, colorectal, and bile duct cancers, particularly in those harboring KRAS mutations, while maintaining low expression in normal tissues. The ADC is designed to exploit the efficient internalization kinetics of TM4SF1 to deliver cytotoxic payloads directly into cancer cells. Preclinical studies conducted by researchers at Yonsei University have demonstrated significant in vitro efficacy and sensitivity in KRAS-mutated solid tumor models, providing a rationale for its development as a precision oncology therapeutic.
