Overview

TMB-365 is a long-acting, broadly neutralizing humanized monoclonal antibody developed as a second-generation version of ibalizumab. It functions as a post-attachment entry inhibitor for HIV-1 by binding to the second domain of the CD4 receptor on host cells, thereby preventing HIV from entering CD4+ cells. Compared to ibalizumab, TMB-365 is designed with improved pharmacokinetics and broader antiviral activity. It is being developed primarily for maintenance therapy in people living with HIV, especially those who are treatment-experienced or require long-term suppression. The drug is administered via intravenous infusion and has been studied both alone and in combination (notably with TMB-380) as part of ultra-long acting regimens intended to improve convenience and adherence for patients[1][2][7][8].
