TMC-310911 (also known as ASC-09) is a novel investigational small molecule protease inhibitor structurally related to darunavir. It was originally developed for the treatment of HIV/AIDS and has demonstrated potent activity against wild-type and multi-protease inhibitor-resistant strains of HIV-1, making it potentially useful for both treatment-naive and experienced patients. The drug acts by inhibiting the HIV protease enzyme, thereby preventing the cleavage of viral Gag and Gag-Pol polyproteins, which results in immature, non-infectious viral particles and blocks viral replication. In addition to its primary indication in HIV infection, TMC-310911 has also been investigated as a potential therapy for COVID-19 due to its ability to inhibit viral proteases from other RNA viruses such as SARS-CoV-2. The drug was developed by Tibotec Pharmaceuticals (now part of Janssen R&D Ireland), but no recent development or approval has been reported[1][2][3][4][5].
