Overview
TMC-647055 is a potent, selective nonnucleoside inhibitor of the hepatitis C virus (HCV) NS5B RNA-dependent RNA polymerase, specifically binding to the NNI-1 site. It is a macrocyclic indole small molecule that disrupts HCV replication by locking the NS5B enzyme in an open, inactive conformation. Developed as an oral, once-daily treatment for chronic HCV infection, it has demonstrated activity against most HCV genotypes and was evaluated in clinical trials up to phase II. The drug was initially developed by Janssen R&D Ireland (formerly Tibotec Pharmaceuticals), with its highest development status now listed as discontinued[3][5][8].
