TMC-649128 is an investigational small molecule antiviral drug that was developed as an oral nucleoside inhibitor targeting the hepatitis C virus (HCV) NS5B RNA-dependent RNA polymerase. Its primary mechanism of action is inhibition of the HCV NS5B polymerase, thereby blocking viral replication. The drug was investigated for the treatment of chronic hepatitis C infection, specifically in genotype 1 HCV-infected patients. Clinical development reached Phase 1 trials but has since been discontinued. The original developers were Medivir AB and Tibotec Pharmaceuticals, with later involvement from Johnson & Johnson[1][3][5][7].
