TMC-Ag3 (also known as Ag3) is a preclinical-stage small molecule therapeutic developed by Arjuna Therapeutics to overcome cisplatin resistance in solid tumors. It belongs to a novel class of drugs called Therapeutic Molecular Clusters (TMCs), which are comprised of clusters of three metallic atoms. TMC-Ag3 acts as a DNA intercalator that disrupts topoisomerase-DNA complex formation and augments chromatin accessibility during DNA replication. Preclinical studies have demonstrated that Ag3 significantly increases the cytotoxic effects of DNA-acting drugs like cisplatin by increasing the amount of cisplatin bound to tumor DNA without affecting normal tissues. These TMCs are small enough to diffuse freely across the blood-brain barrier and are eliminated from the body without being metabolized.
