TMC-Ag5 (also referred to as Ag5) is a preclinical-stage small molecule therapeutic molecular cluster (TMC) being developed by Arjuna Therapeutics for the treatment of KRAS mutant cancers and other solid tumors, including non-small cell lung cancer (NSCLC). The drug belongs to a novel class of therapeutics consisting of five silver atoms arranged in a specific conformation. Its mechanism of action involves the simultaneous and irreversible inhibition of both the glutathione (GSH) and thioredoxin (Trx) antioxidant pathways. By blocking these essential cellular defense systems, TMC-Ag5 induces a massive amplification of reactive oxygen species (ROS), which selectively triggers apoptosis in cancer cells that already exhibit high baseline ROS levels. TMC-Ag5 is orally bioavailable, freely diffusible throughout the body, and capable of crossing the blood-brain barrier, demonstrating significant tumor-killing efficacy in preclinical mouse models with minimal toxicity to normal cells.
