TMC589337 is a small molecule pharmacokinetic enhancer (booster) developed by Tibotec (a subsidiary of Janssen) for the treatment of HIV-1 infection. It belongs to the PEPI (Pharmacokinetic Enhancer Protease Inhibitor) family, which consists of compounds structurally related to protease inhibitors but lacking direct antiviral activity. TMC589337 is designed to inhibit the Cytochrome P450 3A4 (CYP3A4) enzyme, thereby slowing the metabolism and increasing the plasma concentrations of co-administered antiretroviral drugs, such as the protease inhibitor TMC310911. This boosting effect is intended to improve the efficacy and dosing convenience of HIV treatment regimens. The compound was evaluated in Phase I clinical trials to assess its safety, pharmacokinetics, and boosting potential in healthy volunteers.
