Overview

TmCD19-IL18 is an investigational, autologous, 'armored' fourth-generation chimeric antigen receptor (CAR) T-cell therapy being developed by the University of Pennsylvania in collaboration with Kite, a Gilead Company. The therapy is genetically engineered to target the CD19 antigen, which is expressed on the surface of B-cell malignancies, and is further modified to secrete interleukin-18 (IL-18). IL-18 is a pro-inflammatory cytokine intended to enhance the immune response by recruiting and activating additional immune cells within the tumor microenvironment, potentially overcoming resistance mechanisms seen in previous generations of CAR T-cell therapies. TmCD19-IL18 utilizes an accelerated 3-day manufacturing process and is currently being evaluated in a Phase I clinical trial (NCT05989204) for patients with relapsed or refractory CD19-positive non-Hodgkin lymphoma and other B-cell cancers, including those who have progressed after prior CAR T-cell treatments.
