Drug intelligence / Profile preview
tmCLIC1omab is a monoclonal antibody that selectively targets the transmembrane form of Chloride Intracellular Channel 1 (tmCLIC1). Developed by researchers at the University of Milano in collaboration with Primm, tmCLIC1omab is designed to recognize the NH2-terminal domain of CLIC1 that is exposed to the extracellular space when the protein is integrated into the plasma membrane. In solid tumors, particularly glioblastoma (GBM) and colorectal cancer (CRC), tmCLIC1 is chronically expressed on the cell surface of cancer stem cells, where it functions as an active chloride channel. By binding to tmCLIC1, tmCLIC1omab inhibits its chloride channel activity, disrupting the ionic balance required for oncogenic signaling pathways (such as the OSMR/EGFRvIII/STAT3 axis in glioblastoma) and reducing tumor progression. It is being investigated as a potential adjuvant therapeutic and diagnostic (theranostic) tool for glioblastoma and metastatic colorectal cancer.
