Drug intelligence / Profile preview
TMEM175 inhibitors are a class of small molecules designed to block the activity of transmembrane protein 175 (TMEM175), a structurally unique, constitutively active potassium and proton channel localized to the endolysosomal membrane. TMEM175 plays a critical role in maintaining lysosomal pH stability, membrane potential, and autophagic/mitophagic clearance. While TMEM175 loss-of-function mutations are strongly associated with an increased risk of Parkinson's disease, pharmacological inhibition of TMEM175 has been shown to paradoxically accelerate lysosomal macromolecular catabolism and macropinocytosis in certain contexts. Additionally, TMEM175 inhibition has demonstrated therapeutic potential in mitigating inflammatory responses and acute lung injury in macrophages by modulating the ROS-NF-κB signaling axis, and in reducing silica-induced cell death and inflammation.
