TMG-mutTP53 srRNA SA-LNP is an experimental self-replicating mRNA (srRNA) cancer vaccine designed to target 14 hotspot TP53 missense mutations commonly found in pancreatic ductal adenocarcinoma (PDAC). The vaccine utilizes a Venezuelan equine encephalitis virus replicase to achieve high and prolonged protein expression of concatenated p53 mutant peptides. These peptides are encapsulated in sialic acid (SA)-modified lipid nanoparticles (LNPs), which enhance targeting to dendritic cells (DCs). By presenting these TP53 neoantigens, the vaccine stimulates potent and specific anti-tumor CD8+ T cell responses. Preclinical studies have demonstrated its efficacy both as a monotherapy and in combination with anti-PD1 inhibitors in suppressing tumor growth and prolonging survival in PDAC models.
