TML-6 is a novel synthetic curcumin analog developed as an oral drug for the treatment of Alzheimer's disease. It was identified through screening of 12 compounds from Androscience using a platform based on six aging and Alzheimer's disease biomarkers by Professor Ih-Jen Su at Southern Taiwan University. TML-6 exhibits a multi-target mechanism of action, including inhibition of β-amyloid precursor protein synthesis and β-amyloid accumulation, upregulation of Apo E, suppression of NF-kB and mTOR pathways (autophagy activation), anti-inflammatory effects, and increased activity of the anti-oxidative Nrf2 gene. Preclinical studies in two animal models demonstrated its potential to improve or reverse early-stage Alzheimer's disease progression. TML-6 has high bioavailability due to its formulation and has completed preclinical toxicology and safety studies. As of 2024, it is entering Phase 1 clinical trials in the United States with plans for global Phase 2a trials[1][2][4][5][7].
