Tmod logic-gated CAR T-cell therapy is a proprietary cell therapy platform developed by A2 Biotherapeutics for the treatment of solid tumors. The platform utilizes a dual-receptor NOT-gate logic system designed to overcome the challenge of on-target, off-tumor toxicity. Each Tmod product consists of an activating chimeric antigen receptor (CAR) that targets a tumor-associated antigen (such as CEA, MSLN, or EGFR) and a blocker receptor that recognizes HLA-A*02. This system is specifically indicated for patients whose tumors exhibit HLA-A*02 loss of heterozygosity (LOH). In healthy cells expressing both the target antigen and HLA-A*02, the blocker receptor provides an inhibitory signal that overrides the CAR activation, sparing the tissue. In tumor cells that have lost the HLA-A*02 allele, the blocker signal is absent, allowing the CAR to trigger selective T-cell activation and tumor cell destruction. Specific assets within this platform include A2B530, A2B694, and A2B395, which are currently being evaluated in clinical trials such as EVEREST-1 and DENALI-1.
