TMP-301

TMP-301 is a next-generation, orally available small molecule that acts as a selective negative allosteric modulator (NAM) of the metabotropic glutamate receptor 5 (mGluR5). It is being developed to address the neurobiological basis of addiction and has shown promising results in preclinical models for alcohol, cocaine, and opiate use disorders. The drug has demonstrated robust receptor occupancy in humans via PET imaging and an acceptable safety profile in Phase 1 studies. Clinical development includes ongoing Phase 2 trials for alcohol use disorder and planned studies for cocaine use disorder. Originally discovered by Nxera Pharma (formerly Sosei Heptares), it is now being developed by Tempero Bio using structure-based drug design platforms[1][3][4][5][7].
