Overview

TMP-Fc is a recombinant fusion protein, also known as a peptibody, developed by Beijing Tide Pharmaceutical for the treatment of chronic primary immune thrombocytopenia (ITP). It consists of two identical thrombopoietin (TPO) mimetic peptide sequences covalently linked to the Fc fragment of a human IgG1 antibody. TMP-Fc functions as a TPO receptor agonist; it binds to and activates the c-Mpl receptor on the surface of megakaryocytes and their hematopoietic stem cell precursors. This activation stimulates the Janus kinase/signal transducer and activator of transcription (JAK/STAT) signaling pathway, promoting the proliferation and maturation of megakaryocytes, which leads to an increase in platelet production. The Fc domain extends the drug's half-life in circulation via neonatal Fc receptor (FcRn)-mediated recycling, allowing for less frequent dosing compared to native TPO.
