Overview

TMPRSS11E-CCR split-CAR T cells are an experimental, preclinical-stage cellular immunotherapy designed for the treatment of multiple myeloma. This therapy utilizes an 'AND-gate' split-CAR architecture to enhance tumor specificity and mitigate on-target, off-tumor toxicities. The system consists of two separate constructs co-expressed in T cells: a first-generation CAR targeting TNFRSF8 (CD30) containing only the CD3ζ signaling domain, and a chimeric co-stimulatory receptor (CCR) targeting TMPRSS11E containing only the co-stimulatory domain. Potent T cell activation occurs only upon simultaneous binding of both TNFRSF8 and TMPRSS11E on the surface of a tumor cell. Both targets were identified through cell surface proteomics as being highly expressed in multiple myeloma with minimal overlapping expression in vital normal tissues.
