Overview

TmPSMA-02 is a genetically modified autologous chimeric antigen receptor (CAR) T-cell therapy designed to target prostate-specific membrane antigen (PSMA), which is highly expressed in prostate cancer cells. The therapy involves engineering a patient's own T cells to express a CAR that recognizes PSMA, enabling the immune system to selectively attack and destroy cancer cells. This next-generation CAR-T incorporates a CD2 endodomain and additional "armoring" features, including a PD1-CD28 switch fusion protein, intended to enhance efficacy while reducing immune-mediated toxicity and overcoming tumor-induced immunosuppression. Developed initially by the University of Pennsylvania and further advanced by Tmunity Therapeutics (now acquired by Kite Pharma), its primary indication has been metastatic castration-resistant prostate cancer (mCRPC). Clinical development reached Phase 1/2 trials but was discontinued as of August 2023[1][2][3][4][5][6][7].
