TMR4-pro-Talatamab is an investigational tumor-microenvironment-responsive pro-T-cell engager (pro-TCE) designed to treat DLL3-expressing solid tumors, specifically small cell lung cancer (SCLC). Developed by Biocytogen, the molecule utilizes a first-in-class β-glucuronidase-activatable platform. It consists of the bispecific T-cell engager talatamab (which targets DLL3 and CD3) modified with four β-glucuronidase-cleavable masking peptides (TMR4). This masking strategy is designed to prevent non-specific T-cell activation and systemic Cytokine Release Syndrome (CRS), a common dose-limiting toxicity of T-cell engagers. Upon reaching the tumor microenvironment, the masking peptides are cleaved by the enzyme β-glucuronidase, which is typically enriched in tumors, thereby restoring the full cytotoxic activity of the antibody. Preclinical studies have demonstrated that TMR4-pro-Talatamab maintains anti-tumor efficacy equivalent to the unmasked parental antibody while significantly improving systemic safety.
