Overview

TMTP1 is a synthetic tumor-homing peptide consisting of the amino acid sequence NVVRQ (or sometimes reported as RVVRQ), identified using bacterial flagellin library display technology. It specifically binds to highly metastatic cancer cells and micrometastases, including those from prostate, breast, lung, gastric cancers, and hematological malignancies. The primary mechanism involves targeting the cell surface protein XPNPEP2 (aminopeptidase P), which is upregulated in certain cancers and associated with metastasis. TMTP1 has been used as a targeting moiety for diagnostic imaging agents (e.g., radiolabeled or fluorescent probes) and for targeted delivery of chemotherapeutics or cytotoxic proteins to metastatic tumors. It does not have intrinsic anticancer activity but serves as a ligand for tumor-targeted drug delivery systems or imaging agents[1][4][5][6].
