Overview

TMTP1-ICG is a tumor-targeting near-infrared fluorescent probe consisting of the tumor metastasis targeting peptide TMTP1 conjugated to indocyanine green (ICG). It enables specific targeting of tumor tissues for imaging and potential photothermal therapy. The TMTP1 peptide binds specifically to receptors on tumor cells, allowing for enhanced accumulation of ICG in tumors compared to non-targeted formulations. This targeted delivery improves tumor visualization and enables potential therapeutic applications through photothermal effects when irradiated with near-infrared light. ## Composition and Structure TMTP1-ICG is created by coupling the TMTP1 peptide with ICG, often through a PEG4 linker. The TMTP1 peptide has been shown to specifically target tumor cells, particularly cervical cancer cells, while ICG provides fluorescent imaging capabilities. This combination allows for targeted delivery of ICG to tumor tissues, enabling better visualization of tumors and potential therapeutic applications. ## Variants and Formulations Several variants of TMTP1-ICG have been developed: - TMTP1-PEG4-ICG: A linear conjugate with a PEG4 linker between TMTP1 and ICG - (TMTP1-PEG4)2-ICG: A dimeric version with two TMTP1 peptides connected to ICG, showing enhanced tumor targeting ability - TMTP1-modified ICG-loaded polymeric micelles (ITM): Nanoparticle formulation for improved stability and delivery ## Applications The primary applications of TMTP1-ICG include: 1. **Tumor Imaging**: TMTP1-ICG demonstrates excellent tumor-targeting ability, allowing for sensitive detection of tumors, including micrometastases. The (TMTP1-PEG4)2-ICG dimer can identify tumors as small as 10 mm³. 2. **Cervical Cancer Detection**: A fluorescence colposcope using TMTP1-PEG4-ICG has been developed for identifying cervical precancerous lesions, showing comparable results to conventional colposcopy. 3. **Photothermal Therapy**: The ICG component enables photothermal therapy under near-infrared laser irradiation, allowing for targeted tumor treatment. 4. **Lymph Node Mapping**: TMTP1-ICG can be used for sentinel lymph node imaging, particularly tumor-draining sentinel lymph nodes. ## Mechanism of Action TMTP1-ICG works through a dual mechanism: 1. **Targeting**: The TMTP1 peptide specifically binds to receptors on tumor cells, with evidence suggesting XPNPEP2 as a potential target. This allows for active targeting of tumor tissues beyond the passive enhanced permeability and retention (EPR) effect. 2. **Imaging/Therapy**: Once accumulated in tumor tissues, the ICG component provides near-infrared fluorescence for imaging and can generate heat upon laser irradiation for photothermal therapy. The targeting specificity of TMTP1-ICG has been demonstrated in various cancer cell lines, including cervical cancer cells (HeLa, SiHa, C-33a) and breast cancer cells (4T1), with significantly higher uptake compared to non-targeted ICG formulations.
