Overview

tMUC1 CAR T cells are an engineered cell therapy designed to target the tumor-associated form of Mucin 1 (tMUC1), a glycoprotein that is overexpressed and hypoglycosylated in various epithelial cancers, including approximately 80% of pancreatic ductal adenocarcinomas (PDA). The therapy utilizes a chimeric antigen receptor (CAR) incorporating a single-chain variable fragment (scFv) derived from the TAB004 antibody, which specifically recognizes the tMUC1 epitope while avoiding normal MUC1 expressed on healthy tissues. The CAR construct includes CD28 and CD3ζ signaling domains to facilitate T-cell activation, proliferation, and tumor cell lysis upon antigen recognition. Developed by researchers at the University of North Carolina at Charlotte, this approach is being explored as a monotherapy and in combination with low-dose chemotherapy (such as gemcitabine and 5-FU) to enhance efficacy against resistant solid tumors.
