Overview

TMV-018 is a second-generation oncolytic measles virus engineered to encode a potent suicide gene, specifically the prodrug-converting enzyme "super cytosine deaminase" (SCD). Upon intra-tumoral injection, TMV-018 preferentially infects and transfects tumor cells. The expressed SCD enzyme catalyzes the intracellular conversion of the prodrug 5-fluorocytosine (5-FC) into the active chemotherapeutic agent 5-fluorouracil (5-FU), resulting in targeted tumor cell death. This mechanism enhances direct oncolysis and potentiates anti-tumoral immune responses, especially when combined with immune checkpoint inhibitors such as anti-PD-1 antibodies. Preclinical studies have demonstrated significant efficacy and safety in animal models. The drug was developed for use primarily in gastrointestinal cancers and is administered via intra-tumoral injection[1][2][3][4][5][7].
