**TMV-018 + 5-fluorocytosine + anti-PD-1 inhibitor** is a combination therapy under clinical evaluation for the treatment of solid tumors, particularly gastrointestinal tract cancers. TMV-018 is a second-generation, measles vaccine-based oncolytic virus engineered with a suicide gene that encodes cytosine deaminase. Upon administration of the pro-drug 5-fluorocytosine (5-FC), cytosine deaminase converts 5-FC into the chemotherapeutic agent 5-fluorouracil (5-FU) within tumor cells, enhancing tumor cell lysis (oncolytic effect). The therapeutic effect can be further potentiated by concurrent administration of an anti-PD-1 inhibitor, which acts as an immune checkpoint inhibitor to enhance antitumor immune responses. This multi-component therapy aims to combine direct oncolysis, localized chemotherapy, and immune checkpoint blockade for synergistic anticancer activity[1][3][5].
