TN-301 is a highly selective, first-in-class small molecule inhibitor of histone deacetylase 6 (HDAC6) developed by Tenaya Therapeutics. It is being developed primarily for the potential treatment of heart failure with preserved ejection fraction (HFpEF), a form of heart failure characterized by diastolic dysfunction, inflammation, fibrosis, and metabolic defects. TN-301 has a multi-modal mechanism of action that includes modifying cytoskeletal and other proteins to coordinate cellular processes. Preclinical studies have shown that TN-301 can reverse many signs and symptoms of HFpEF, improving cardiac function, glucose tolerance, and reducing inflammation and fibrosis. Phase 1 clinical trials demonstrated safety, tolerability across a broad dose range with no serious adverse events or dose-limiting toxicities observed. Pharmacokinetics showed dose proportionality supportive of once-daily oral dosing with robust target engagement confirmed by pharmacodynamic markers such as tubulin acetylation.
