TN01 series is a group of small molecule inhibitors targeting Fat Mass and Obesity-associated protein (FTO), an RNA demethylase. Developed by Tinos Therapeutics, these compounds are designed to modulate RNA methylation (specifically m6A) to treat various cancers. FTO is often overexpressed in certain malignancies, where it promotes oncogenesis by demethylating m6A on specific mRNA transcripts, thereby enhancing the stability or translation of oncogenes. By inhibiting FTO, the TN01 series aims to restore normal m6A levels and suppress tumor growth. The program is currently in the preclinical (Pre-IND) stage for indications including glioblastoma, colorectal cancer, and acute myeloid leukemia (AML).
