Drug intelligence / Profile preview
TN13 (also known as TAT-TN13 when conjugated with a cell-penetrating peptide) is a first-in-class peptide-based p38 mitogen-activated protein kinase (MAPK) inhibitor, specifically targeting the p38α isoform (MAPK14). Originally discovered by the Korea Research Institute of Bioscience and Biotechnology (KRIBB) and developed by Ingenium Therapeutics, TN13 is derived from the docking motif of thioredoxin-interacting protein (TXNIP) and inhibits the TXNIP-p38 interaction. In oncology, TN13 is being evaluated as a payload to restore natural killer (NK) cell function by protecting them from the suppressive effects of tumor-secreted TGF-β within the tumor microenvironment. It is being developed in combination with Ingenium's memory NK cell therapy, gengleucel (IGNK001), for solid tumors such as lung cancer. Additionally, preclinical studies have demonstrated its therapeutic potential in acute inflammatory conditions, including acute respiratory distress syndrome (ARDS) and sepsis, as well as in osteoporosis and hematopoietic stem cell rejuvenation.
