TN4510 is a novel dual-target inhibitor identified through drug screening, designed to address intrinsic resistance to neoadjuvant chemoimmunotherapy in lung adenocarcinoma (LUAD) patients. It functions by binding to both the CHCHD3-S2 domain of Coiled-coil-helix-coiled-coil-helix domain-containing 3 (CHCHD3) and the cGAS-S3 domain of cyclic GMP-AMP synthase (cGAS). This dual inhibition aims to counteract CHCHD3-mediated suppression of the cGAS-STING pathway, thereby restoring antitumor immunity and enhancing the efficacy of neoadjuvant therapy. Preclinical studies in humanized NSG mice have shown that combining TN4510 with neoadjuvant immunotherapy synergistically inhibits tumor growth.
