TNF-α preconditioned mesenchymal stem cells (TNF-α-MSCs) are an experimental cell therapy product consisting of mesenchymal stem cells that have been exposed to tumor necrosis factor-alpha (TNF-α) ex vivo to enhance their therapeutic potential. This preconditioning 'primes' the MSCs, significantly boosting their immunomodulatory capabilities compared to naive MSCs. In preclinical models of atherosclerosis, systemic administration of TNF-α-MSCs has been shown to promote the expansion of regulatory T cells (Tregs), increase the production of the anti-inflammatory cytokine IL-10, and suppress pro-inflammatory cytokines such as IFN-γ and TNF-α. These effects contribute to a reduction in atherosclerotic lesion size and an improvement in systemic lipid profiles, including decreased LDL and increased HDL levels.
