TNF AcTakine is a research-stage immunocytokine belonging to the novel class of 'Activity-on-Target cytokines' (AcTakines), developed by Orionis Biosciences in collaboration with VIB and Ghent University. It is a recombinant fusion protein consisting of a mutant form of Tumor Necrosis Factor (TNF) with significantly reduced affinity for its receptors, fused to a single-domain antibody (sdAb or nanobody) that targets CD13 (aminopeptidase N), a marker specifically expressed on tumor neovasculature. This design allows for the selective delivery and activation of TNF signaling (primarily via TNF-R1) on tumor endothelial cells, thereby inducing vascular disruption and anti-tumor effects while avoiding the severe systemic toxicity associated with wild-type TNF. Preclinical studies have demonstrated its potential in treating solid tumors such as melanoma and lung carcinoma, particularly when used in synergistic combinations with other targeted cytokines like IL-1β AcTakines.
