This is a monoclonal antibody targeting tumor necrosis factor-alpha (TNF-α) being investigated by Shanghai Changzheng Hospital for the treatment of traumatic acute spinal cord injury (SCI). TNF-α is a key pro-inflammatory cytokine that drives the secondary injury cascade following the initial trauma to the spinal cord, leading to ischemia, apoptosis, and further functional loss. By inhibiting TNF-α shortly after injury (within the acute phase of 2 weeks), the therapy aims to mitigate this inflammatory response and improve neurological recovery. The drug is administered via subcutaneous injection (50mg) and is being evaluated in a Phase 1/2 clinical trial (NCT04988425) comparing its safety and efficacy against methylprednisolone and saline. While TNF-α inhibitors are established treatments for autoimmune conditions like rheumatoid arthritis and ankylosing spondylitis, their application in acute SCI represents a novel neuroprotective strategy.
