Overview

TNF-T + cisplatin + gemcitabine is a combination therapy consisting of three agents: - **TNF-T** (presumed to be a form or derivative of tumor necrosis factor alpha, TNF-α): TNF-α is a cytokine with direct anti-tumor and antivascular effects. It can increase the permeability of tumor vasculature, thereby enhancing the delivery and efficacy of chemotherapeutic agents such as cisplatin and gemcitabine. While systemic use has been limited by toxicity, regional administration (e.g., isolated limb perfusion) has shown synergistic effects with chemotherapy[6][9]. - **Cisplatin**: A platinum-based alkylating agent that cross-links DNA strands in cancer cells, leading to apoptosis. It is widely used in various solid tumors[7][5]. - **Gemcitabine**: A nucleoside analog that inhibits DNA synthesis by incorporation into DNA during replication, resulting in cell death. It is commonly combined with cisplatin for enhanced antitumor activity[1][3][5]. The rationale for this combination lies in the potential synergy between TNF-mediated vascular disruption (increasing drug penetration) and the cytotoxic actions of both chemotherapeutics.
