TNFAIP3 siRNA is a small interfering RNA designed to silence the expression of the TNFAIP3 (Tumor Necrosis Factor Alpha-Induced Protein 3) gene, which encodes the A20 protein. A20 is a potent zinc-finger protein that serves as a critical negative regulator of the NF-κB signaling pathway, helping to terminate inflammatory responses. In oncology research, particularly concerning gliomas and glioblastomas, TNFAIP3 siRNA is utilized as a tool to investigate the molecular mechanisms of resistance to alkylating agents like temozolomide (TMZ). Knockdown of TNFAIP3 leads to the constitutive activation of NF-κB, which in turn upregulates anti-apoptotic proteins such as Bcl-2 and BIRC3, conferring resistance to TNF-alpha and TRAIL-induced apoptosis. Research indicates that TNFAIP3 is a direct target of microRNAs miR-125a and miR-125b, and siRNA-mediated inhibition of TNFAIP3 mimics the phenotype of miR-125a/b overexpression in promoting tumor cell survival and therapy resistance.
