Drug intelligence / Profile preview
TNFR2 agonistic antibodies are a class of monoclonal antibodies designed to bind and activate tumor necrosis factor receptor superfamily member 1B (TNFR2/TNFRSF1B). TNFR2 is highly expressed on regulatory T cells (Tregs), myeloid-derived suppressor cells (MDSCs), and certain endothelial and neuronal cells. In the context of autoimmune and inflammatory diseases, such as type 1 diabetes and multiple sclerosis, TNFR2 agonism is utilized to selectively expand and stabilize highly suppressive regulatory T cells (Tregs), thereby restoring immune tolerance and suppressing autoimmune responses. Conversely, in oncology, certain TNFR2 agonistic antibodies (such as BI-1910 and HFB200301) are being developed to stimulate CD8+ effector T cells and natural killer (NK) cells to enhance anti-tumor immunity, or to deplete tumor-infiltrating Tregs via Fc-mediated effector functions.
