TNFRSF8 2nd generation CAR T cells are an experimental chimeric antigen receptor (CAR) T cell therapy designed to target the TNFRSF8 (CD30) antigen. Described in preclinical research by investigators at the University of Cambridge, this construct utilizes a standard second-generation CAR architecture, which typically incorporates an extracellular antigen-binding domain (scFv) linked to an intracellular CD3-zeta signaling domain and a co-stimulatory domain (such as 4-1BB or CD28). In studies focused on multiple myeloma, this specific construct was used as a benchmark to evaluate the sensitivity and specificity of novel "AND-gate" split-CAR systems. Research presented at ASH 2023 indicated that while the 2nd generation TNFRSF8 CAR was insufficient to trigger activation against cells with low antigen expression, it serves as a foundational tool for investigating combinatorial antigen recognition strategies in hematologic malignancies.
