TNFRSF8-CAR is a chimeric antigen receptor (CAR) T-cell therapy designed to target TNFRSF8, more commonly known as CD30. CD30 is a cell surface marker belonging to the tumor necrosis factor receptor superfamily, which is highly expressed in several hematologic malignancies, including Hodgkin lymphoma (HL) and systemic anaplastic large cell lymphoma (ALCL), while having limited expression in healthy tissues. The therapy involves the ex vivo genetic modification of autologous T cells to express a CAR construct typically comprising an anti-CD30 single-chain variable fragment (scFv) linked to intracellular signaling domains such as CD28 or 4-1BB and CD3-zeta. Upon reinfusion, these engineered T cells recognize and eliminate CD30-expressing tumor cells. This approach has been primarily investigated by researchers at the National Cancer Institute (NCI), led by Dr. James Kochenderfer, for the treatment of relapsed or refractory CD30+ lymphomas and potentially multiple myeloma.
