TNFRSF8 CAR-only T cells are an experimental cellular therapy component developed by researchers at the University of Cambridge for the treatment of multiple myeloma. These T cells are engineered to express a first-generation chimeric antigen receptor (CAR) targeting TNFRSF8 (also known as CD30). This specific construct contains an intracellular signaling domain (typically CD3ζ) but lacks a co-stimulatory domain (such as 4-1BB or CD28). In the context of a split-CAR or AND-gate system, these cells are designed to be used in conjunction with a second construct targeting a different antigen (e.g., TMPRSS11E) that provides the necessary co-stimulation. On their own, TNFRSF8 CAR-only T cells are intended to be non-functional or exhibit minimal activity to prevent off-tumor toxicity against tissues expressing low levels of TNFRSF8, requiring the presence of both targets for full T-cell activation and tumor cell lysis.
